I wouldn't think so unless you shut off the engine which would stop fluid pumping through the transmission. That's why you shouldn't tow a vehicle with an automatic very far with the drive wheels on the ground. I believe there are a few cars out there with fluid pumps on the output end of the transmission which would be ok.
